Immunohistochemistry (IHC):
AE00234 to STAT3-pS727 was successfully used to stain nuclei in epithelial cells of human, mouse and rat tissue sections. Recommended concentration: 5-15ug/ml
Formaldehyde-fixed, paraffin-embedded human lung (left) and breast carcinoma (right) stained with STAT3-pS727 Rabbit Phospho-specific Polyclonal Antibody AE00234 at 10ug/ml for 30 minutes at RT. Epitope retrieval: Microwave heating at pH7 to boiling, 5min at 95-98C, 1min at 100C, then cool to roomtemp. DAB staining by HRP polymer.

Immunoprecipitation (IP):
AE00234 to STAT3-pS727 was successfully used to immunoprecipitate an approx. 110kDa band from lysates of serum-starved and subsequently PMA/TPA-stimulated HeLa. Recommended amount: 3-6ug
Western Blot of immunoprecipitation (lanes 2 and 3) from 200ug HeLa (PMA/TPA at 200nM, 30 min after overnight serum-starvation) extract, using 3ug STAT3-pS727 Rabbit Polyclonal Antibody AE00234, and using the same antibody for staining the blot at 1ug/ml (1h at ambient temp). ECL staining by HRP.
Western Blot (WB):
AE00234 to STAT3-pS727 was successfully used to stain an approx. 90kDa band in lysates of NIH3T3 and C6 after anisomycin or UV-treatment. Recommended concentration: 1-3ug/ml
Western Blots of 30ug lysates of NIH3T3 and C6 with and without Anisomycin (25ng/ml, 30 and 20 min resp), or UV (15-30min)-radiated, stained with STAT3-pS727 Rabbit Polyclonal Antibody AE00234 at 1ug/ml (1h at ambient temp). ECL staining by HRP.
